I spent a 24 hour layover in Philly with the gf a weekend or two back. We didn't get much of a feel for the city and the frigid weather prevented us from doing the tourist thing, but it was still a fun little jaunt. Got to wander around Reading Market and hit a couple of local bars/restaurants within striking distance of the train station.
