Inspect the inside and underside of all belts and hoses for fraying and cracks. High use and high temperatures often break less than perfect bets and hoses.
Test all lights, blinkers and turn signals. Not only for safety, but there is no reason to attract police officers in every town you pass through.
Check levels of engine oil, coolant, brake fluid and power steering fluid.
Inflate your tires to the proper pressure. Confirm that they have good tread, with no cuts, imbedded items or bulges. This will not only cut down your chances of a blowout or flat, it will also improve gas mileage significantly.
Pack an emergency kit specifically for the trip that includes water, motor oil, flares, jumper cables and tools for removing a tire and basic work on the car. You should also have a emergency medical kit in your car.
Related, but not exactly what you're talking about: make sure your registration is up to date, with any paperwork for that (and your proof of insurance) in your glove compartment. Any lapsed stickers on your car will put you on the grid. No sense in causing yourself trouble over something you can correct before you go!
Inflate tires to correct pressure, you'll get better gas mileage this way. As, if you need an oil change, get one. Change your air filter (you can do this yourself with 1 screwdriver).
I've got a not so nice little vibration in my front end that comes and goes at around 70mph. Me thinks it's an alignment issue from a pot hole, hoping to get it take care of this weekend. My oil will be due mid-trip, so I'll just eck that out as well.

DON'T inflate your tires to the pressure on the tire, look in your manual or on the inside of your door and that will tell you the correct pressure. The number on the tire is it's maximum inflation and this will cause you to have worse gas mileage and a much better chance at getting a flat!
